We hear a lot about how TSH isn't a very reliable marker for thyroid disease...but what about TRH (thyroid releasing hormone) that stimulates TSH in the first place?

It turns out that TRH does a whole heck of a lot more than simply stimulates the thyroid gland. It appears quite intimately involved in a number of CNS activities and has potential for use in many diseases including depression, OCD and autism as a neuromodulatory agent.

It can help modulate acetylcholine (the hormone of the vagus nerve) and NE, improve memory, increase dopamine, and is a novel pain reliever.

It seems logical that there would be potential in ME/CFS as well.

Oral, sublingual tablets are available in other countries (some even available online) but are quite expensive at this point. I would love to see some research studies so these drugs could potentially have wider use in this country too.

Taltirelin's effects on NEUROTRANSMISSION

  • Taltirelin increases extracellular levels of acetylcholine in the hippocampus, but modulates turnover if levels are already high (potentially protecting against excesses of ACh as well).
  • Taltirelin increases noradrenaline in the hypothalamus.
  • Taltirelin super-potently increases dopamine release in the striatum and nucleus accumbens - which are area's involved in motivation and as such this is likely the responsible action for it's pungent anti-depressant effects.
  • The compound also can regulate/increase serotonin levels in the brain stem. nucleus accumbens and striatum.
  • Taltirelin may increase neurite outgrowth via distinct modulation of neurotrophic factors and via positive effects on blood flow.

In summary,
TALTIRELIN, marketed as Ceredist and often referred to as TA-0910, has demonstrated the following effects/benefits.
  • As an anti-depressant(!)(!).
  • As a nootropic(!).
  • Ameliorating memory deficits, and increasing retention(!).
  • As a novel analgesic/pain reliever(!).
  • As a stimulant - with motivation increasing effects(!) (!).
  • Regenerating spinal cord nerves and treating spinal muscular atrophy(!).
  • Rapidly increasing dopamine, noradrenaline, serotonin and acetylcholine(!) (!).
  • Amelioration of Autism Spectrum Diorder (ASD) and Obsessive-Compulsive Disorder (OCD) conditions.

With input from @Hip, it looks like dosing ranges from as little as 500 mcg up to 2mg/day. Unfortunately, there doesn't seem to be a lot of consensus on dosing, especially for neuro conditions.

TRH was also mentioned by Dr Goldstein in Tuning the Brain...both as a 500 mcg IV injection (given once every few days) and a very low dose nasal spray delivering only about 3 mcg used 3x/day.

It looks like the higher doses may be better for depression type illnesses than the lower doses but again the research is slim.

Another version here..."proprietary", so not sure how it compares to the one from Peptide Sciences above, but it is the same as the ODT Ceredist from Japan, supposedly.

TAO-OA future trials within human clinical settings, and based off of the animal studies thus far so conducted, would appear to indicate optimized dose arms of 1mg and 2mg once per day or every other day per oral and intranasal administration, and 0.2mg and 0.4mg once per day or every other day per s.c. injectable administration.

These doses appear to be applicable for antidepressant effect, arousal effects, pro-metabolic effects, cognitive enhancement effects, anti-aging effects, as well as for several other benefits. Notably, within potential usage for autism, and as well severe disorders of other natures, a dose of ~2-3x that as indicated would seem to be most warranted to be assessed within clinical exploration and testing.

I think this TAO-OA product is a mixture of taltirelin and regular thyrotropin-releasing hormone (aka protirelin).
