Treatment of Activated Microglia with Ibudilast, Minocycline & Amitriptyline

i found some interesting stuff in one of my books, it says that Microglia gets activated by systemic immune modulation (e.g. interferon) and stays in this state forever. the microglia then produces ROS, inflammatory cytokines and so on which encounters the nerves and leads to cell apoptosis (cell death), this can get modulated by Minocyclin, which depolarizes Microglia and inhibits the inflammation state. Very selective!

Is anyone out here that has experience with Minocycline?

And anyone who ever tried out Ibudilast? That also acts at the root of cause, Microglia activity.

In combination with a low dose of Amitriptylin and Curcumin, both Anti inflammatory in the CNS, this will be my next experiment. With and without Prednisone. Is anyone out here that has experience with Minocycline?

Look into memantine. Ive had some success with it. It has anti inflammatory effects against several cytokines, microglial and is an nmda antagonist. I only every used low dose 5 to 10mg but did help energy some.

Aren't most of the ME/CFS specialists who treat microglial activation using LDN?

ok, i got a script for Minocyclin and it makes me really calm down.... so down that i can lay on the bed and just listen to music without thinking that i get stabbed or sth like that... it really does the trick! I hope this is not placebo related and keep you updated on the minocyclin front......

wooooooow... so i didn't take Prednisone hours ago and i STILL have no exhaustion from loud music (i hear metal mostly) and from playing fast riffs on the guitar... this is the first time in over 3 years that that's possible... i still think minocyclin can handle a lot of the inflammation circle on its own, without the help of cortisone or what else... i've played over 30min, fast ,with a rest phase of 30min, then again, 30min playing without loosing energy... this minocyclin is reallypowerfull stuff... i pray to god that it's not placebo:nurse:

Hope you'll keep us posted, Loki. I've been very interested in minocycline for awhile, but my neurologist wouldn't prescribe it, so I put the idea on hold. (Memantine hasn't worked well for me.)

What dosage are you using? Do you expect to use it short-term only?

Hope it continues to work well for you.

thank you! well, i got a pretty big box (100 pills) 50mg and i take 200mg per day. 1 every 6hours.
it makes you more calm than a blister of Ativan and i'm in love with this stuff!:D

the thing is, i think to reset the microglia (when that's possible...) treatment should take some months to a year. the whole inflammation thing is driven by astroglia and microglia and their reactiveness. when you inhibit microglia, astroglia will also calm down because it's dependent on microglia response chemicals. that's a feedback loop. Mino seems to cause this feedback loop to interrupt and when i remember correctly there is a chance of getting the glia back to sleep after some time of treatment.
i hope i'm right! because this could help many people i guess?! at least i don't need so much Prednisone anymore:smuggrin:

stupid that it can't undo the damage already done but i stick with it as long as i don't throw up all the time or my nails turn purple ... it saves me from sexually transmitted diseases, i'm okay withthis too.
i'm gettingalittleparanoid about the things that happenat the moment, maybe i get more crazy and die ! i hope that i die someday, but first i want to do some things i always wanted to do, the point is, will i lose everything? Will it get better? stay tuned on the minocyclin show

So are you saying the minocycline is now giving you a wild ride? Or something else? How's the guitar playing going?

yes, i got prescribed interferon-alpha in 2014 and it made me a human vegetable. well... i now can play fast and easy like back in the days (on e guitar) with Minocyclin. I don't know why, i still feel like i lost 50% of my IQ but at least i can play fluidly (without any feelings though).

maybe that's the lyrica. i doNt# feel nothing. No thing. but canplay like a boss. i don't understand that... why? maybe it's from the lyrica... SOMETImeS IT'S REALLY HARD IN LIFE. YOU FALL DOWN AND ENDLESSLY FALL and nobody stops that fall I WOULD RATHER BE INSANE, BBUT IT'S ALL THIS RAGE ,aybe i should kill myself, back when that was actual , there is no understanding of the situations aand when they get understandable you cannot be happy about that. there is no way backwards, but also not forwards, when there is a frorward
